My PS-100 doesn’t work normally, since I misconnected the amp in and the speaker out. I measured the plug of the output transformer and found it didn’t have resistance(short circuit).
So is the output transformer broken? Or it’s something else.

A bit more information about the current state would be useful.
With just, guitar, amp, PS-100 and speaker connected (no FX) does the PS-100 work in bypass? Does it work in operate?
The DC resistance of the output transformer is very low, so it’s not really reliable way to diagnose faults (unless you really know what you are doing and have a good equipment etc.)
How long did you run the amp into the SPKR OUT? This is not good for the circuit but tubes are pretty robust and might be fine for a short time.
